I've got new R600 levers with single button hoods, but an
older SM-6500 bracket/sensor kit, made for the two-button
right hood. What is recommended: shall I get a new
bracket/sensor kit for single-button hoods, or shall I
instead try to fit a two-button hood on the R600 lever?

Neither. Just install it and ride.
The "buttons" in the hoods are not functional, they
just serve as a tactile indicator of where the actual
buttons are.
Before I switched to Campagnolo, my Hetchins had 105
brifters with the both-buttons-on-the-right rubber hoods,
but with the one-button-on-each-side hardware, worked
just fine.
