I'm creating a video for a large cycling tour (around 3,000 riders). I going to interview a few riders who have rode it many times. As a rider what questions would you find interesting, entertaining or informative to hear the answer to?

I would have thought, by looking at your username, that you would have no trouble of thinking up some questions ;)
What questions would you like to question folk about...
Questionables:
The course: start and finish locations - with things to do at either location on the day of the event. Items of interest enroute. Difficulties on the course - if there's been previous tours or similar events in the area what challenges have there been? This could be influenced by terrain or weather - or both. Maybe work in something about what the riders think of amenities provided if there are any or oddball things that the organizers do - the Death Ride has the "angels" on the top of the highest pass, the Alta Alpina Challenge has the Tiki Lounge on the final pass
The location: Include stuff to do before and after the event - this could also be useful for riders are bringing along a significant other that will not be riding. Places to eat, good hotels - especially ones that go out their way to cater for cyclists - local attractions etc. Find out what other riders have done in prior years.
If there are some places that are frequently recommended go see if they can put on some deal for the event or sponsorship in return for a spot on the video...

Here's a few that come to my mind. I tend to think of the best interviews as being simply conversations.
- What got you started in cycling? How has it driven you to engage in this XXX mile event?
- How long have you ridden? Ridden seriously?
- What's more important a bike that looks great, rides great, or performs great?
- Now that you've completed this tour, what would be your fantasy tour?
- Does anyone in your family consider you to be slightly crazy? Although lovable?
