How tight should I screw the roller against the tyre? It says tight enough to avoid tyre slippage, but this is still quite a grey area. It also warns against "too tight".

I normally tighten mine about 2 and 1/4 to 2 and 1/2 full turns, seems to do the trick for the tires I use (Cheap Bontangers),
Tighten it enough so that when you quickly pull the tire by hand, and then reverse direction, there is no slip.
If you are really keen, you can do a rundown test. I've read that some people speed it up to 20 mph, then let it coast down to 0. It should take, if I believe what I've read, 13.4 seconds. No idea where that number comes from.
On the other hand, a rundown test, regardless of the number, will let you have the same tension on the tire next year as this year, which could be useful if you like to do comparisons.
