    This would give some credence to the theory of targeting of iron as an antiinflammatory. Since rutin
    is a potent iron binder.

    In the present investigation we have evaluated the potential existence of analgesic activity of the
    residue of tincture and of the crude lipophilic (AC 2, AC 4) and hydrophilic portions (AC 1, AC 3)
    of infusion and decoction obtained from flower heads of Achillea collina J. Becker ex Reichenb. Acid
    acetic induced writhing and vascular permeability test was performed by the method of Whittle.
    Comparison was made with indomethacin, morphine and rutin, as references drugs. The mice were dosed
    orally with samples. AC 2 induced a potent inhibition (47.23%; p < 0.05) of the nociceptive response
    after oral administration of 250 mg/kg--a dose ten folds higher than the dose of indomethacin. AC 3,
    at a dose of 5000 mg/kg (p.o.) inhibited with 47.6% (p < .001) and 19.8% (p < 0.05) the increase in
    peritoneal vascular permeability caused by 0.5% acetic acid. Relative unexpected, morphine (1.14
    mg/kg, p.o.), an opioid analgesic, also produced a significant reduction of the leakage
    (41.2%, p < 0.01). This finding is consistent with the reports in which the role of the endogenous
    opiate system in the physiological response to injury/inflammation, mainly controlling fluid
    leakage into the interstitial space, is sustained.

    Anti-inflammatory effect of rutin on rat paw oedema, and on neutrophils chemotaxis and
    degranulation.

    BACKGROUND: Rutin, a natural flavone derivative, is known for its pharmacological properties. We
    have previously reported that this flavonol exerted a potent inhibitory effect on respiratory burst
    of fMet-Leu-Phe-stimulated neutrophils, as well as on phosphoinositide 3-kinase gamma activity in a
    cell free system. In the present study, the anti-inflammatory effect of rutin was investigated in
    vivo and in vitro. METHODS: rutin or aspirin (100 mg/kg, body weight) were given orally to rats 1
    hour before paw oedema induction, using lambda-carrageenan 1%. The rat paw volume was measured by
    mean of plethysmometer, initially and during 6 hours. The chemotaxis of neutrophils towards 10(-7) M
    fMet-Leu-Phe was performed using 48-well chemotaxis chamber. Neutrophils that migrated through 5
    microm pore size polycarbonate filter, in presence or in absence of rutin, were counted
    microscopically. Elastase exocytosis of either phorbol 12-myristate 13-acetate or fMet-Leu-
    Phe/cytochalasin B-stimulated neutrophils was assessed in absence or in presence of rutin using the
    synthetic substrate N-Suc-Ala-Ala-Ala-p-nitroanilide. The absorbance of released p-nitroaniline was
    measured at 405 nm using microplate reader. RESULTS: The maximal swelling in placebo group was
    observed at 5 hours, after lambda-carrageenan injection. Oral administration of rutin reduced rat
    paw swelling starting 2 hours after lambda-carrageenan injection. Rutin reduced significantly (p <
    0.05) and in a dose-dependant manner the polymorphonuclear neutrophils chemotaxis to fMet-Leu-Phe.
    Furthermore, elastase exocytosis, induced by both stimuli, was partially inhibited by rutin up to 25
    microM. CONCLUSION: The present study revealed that rutin possesses anti-inflammatory properties.

    Oxidative stress in rheumatoid arthritis leukocytes: suppression by rutin and other antioxidants and
    chelators.

    The enhanced production of superoxide ion and peroxynitrite by bloodstream neutrophils and of
    superoxide ion by monocytes from rheumatoid arthritis (RA) patients was registered. It was suggested
    that NADPH oxidase together with NO synthase were the major sources of superoxide ion in RA
    neutrophils, while in RA monocytes superoxide ion was produced by NADPH oxidase and mitochondria.
    Among the different free radical inhibitors studied (antioxidant enzymes, SOD and catalase; free
    radical scavengers, bioflavonoid rutin and mannitol; and the iron chelator desferrioxamine), SOD and
    rutin were the most efficient suppressors of oxygen radical overproduction by RA neutrophils, while
    mannitol and desferrioxamine were inactive. Thus, in contrast to Fanconi anemia (FA) leukocytes
    (Korkina LG et al., J Leukocyte Biol 1992;52:357-62), iron-catalyzed hydroxyl radical formation was
    unimportant in RA leukocytes, which mainly produced superoxide ion. Natural non-toxic bioflavonoid
    rutin (vitamin P) inhibited oxygen radical overproduction in both RA and FA in an equally efficient
    manner and therefore may be considered as a useful supporting pharmaceutical agent for the treatment
    of "free radical" pathologies.

    Different antioxidant activities of bioflavonoid rutin in normal and iron-overloading rats.

    The effects of rutin on liver microsomes, peritoneal macrophages, and blood neutrophils isolated
    from iron-overloading (IOL) and normal rats were studied. The formation of 2-thiobarbituric acid-
    reactive products and the level of lucigenin-amplified chemiluminescence (CL) were determined in
    liver microsomes. Oxygen radical production by phagocytes was measured by luminol- and lucigenin-
    amplified CL and superoxide dismutase-sensitive cytochrome c reduction. These ex vivo findings were
    compared with the in vitro effects of rutin on cellular free processes. It was found that rutin
    administration sharply suppressed free radical production in liver microsomes and by phagocytes of
    IOL animals and only slightly affected these processes in normal rats. The selective inhibitory
    effect of rutin under pathologic conditions induced by iron overload is thought to be due to the
    formation of inactive iron-rutin complexes which are unable to catalyse the conversion of superoxide
    ion into reactive hydroxyl radicals, a process responsible for the free radical-mediated toxic
    effects of iron overload. These findings may account for the favourable effects of the treatment of
    pathologies associated with iron overload with rutin.
