You don't know and you don't need to know, if it's only a tiny puncture
and the stuff worked.
I've used WrenchForce tubes that contained a sealant liquid. Larger
punctures were obvious from a hissing or bubling sound/sight. These
punctures are left in the 6 o'clock position to give the stuff a chance to
work. Doesn't always work, sometimes requires patching as normal. In
fact it's a good idea to patch anyway, eventually, if you find what look
like punctures in the tube.

Not really. The stuff sloshes about evenly, I suppose. Weight is a
factor, though: much heavier than normal tubes

Maybe if you're terribly bothered by punctures -- either just the
frequency or you /really/ don't like fixing them, ever. Highly puncture
resistant tyres is normally a good enough solution, IMO.
