Well, that's possible-- and I have even done it before with disk brake
hubs and gearhubs. I still have specially made 72 hole Velocity Deep-V
rims if I want to use those. However, not only is that a lot of
painstaking work (54 holes must be precisely located and drilled), but
I don't think it would result in a significantly stronger wheel than a
substantially heavier rim laced with 36 spokes.
On the plus side, the Deep-V with 72 spokes would be lighter and
cooler-looking. On the minus side, it would not fit very well with the
wide tires I intend to use.

My current electric bike, using a nominal 400W scooter motor and
weighing about 70 lbs., uses a 36 hole BMX hub with a Sun Rhyno Lite
700c rim on the rear. The wheel requires more frequent truing than my
pedal bikes' wheels (like every 100 miles or so), but seems reasonably
well-suited to the task.
The hub motor I would like to use is a 750W unit in a front wheel. The
hubmotor alone weighs about 12 lbs. The bike will be capable of higher
speeds than the one I currently use, and may be heavier due to a larger
battery pack. On the other hand, only the front wheel will have 36
spokes-- the rear will have 48.
